Margaret Hughes, playing Desdemona in Othello in 1660, is believed to be the first professional woman to perform on the public stage in England. In William Shakespeare's time all female roles were taken by boys whose voices had not yet broken, or by older men in the players' company. Only in private performances, such as masques at court were women allowed to act on stage, and then only alongside professional players.
The notorious female transvestite Mary Frith is said to have taken part in a play at the Fortune Theatre ca. 1611, but it would not have been one of Shakespeare's. (It may have been the Roaring Girl by Middleton and Dekker, which was actually written about her)
Women appeared in Shakespeare's plays from the very start. He wrote no plays that had no female parts (although Julius Caesar comes close). However, those female parts were acted by men and boys until 1660, when the female parts were played by actresses for the first time.
Women did appear on the Elizabethan stage at least occasionally. Mary Frith, the famous Elizabethan cross-dresser, was charged with having appeared on the stage of the Fortune Theatre while singing a song. That would not have been during a Shakespeare play, however, since the Fortune belonged to the competition to the King's Men.

Are you talking about at the time of Shakespear? Because it was unseemly for women to display themselves in such a manner in public. Kissing went on in the plays, and no woman kissed anyone other than her husband. 'Acting' wasn't a consideration.

The first time a woman acted in a Shakespeare play was in 1660. The production was Othello, and the actress played Desdemona on December 8. There is some dispute as to her name, but most think it was Margaret Hughes.

My guess is Empress Zewditu of Ethiopia who appeared on a 1918 stamp of Ethiopia. You didn't ask from what country, so it's hard to say. If you are talking about the United States, the first Black woman to appear on a postage stamp was Harriet Tubman on Feb 1, 1978.
