The president's term expires after four years. At time, the voters can replace him if they wish and by law he can serve only two terms, so he is sure to be replaced after 8 years.
If the president is unable to perform his duties or resigns, the vice-president replaces him
There is also a lengthy process by which Congress can remove a President from office and replace him by the vice-president.

If a vice president takes office after his president has served no more than half his term, then the vice president can finish the president's term, anywhere under two years. Then this president can serve the two term limit on his own, or a possible total of ten years. However, this has never occurred.

The 25th Amendment deals with death or incapacitation of the chief executive officer i.e., President or Vice President.
When a President dies in office, the Vice President automatically replaces him. There is no election until he finishes the deceast President's term of office. So, I'm not sure if that is what you intended to ask.
The vice president would finish the term and have to run for any future terms on his own. Eight years (two terms) is the maximum time anyone can be president.
