According to the National Archives, women may have fought in the civil war, though they disguised themselves as men. A statement from the Army, which you will find in the attached link, says the Army basically denies and knowledge of females fighting in the civil war, but that some females could have fought without their gender having been identified.

In wars in the United States, there have been two types of solders: regular army solders, and local militias. During the Revolutionary War, a few teen age girls disguised themselves as boys and joined the regular army. In more cases more women joined in local militias when the enemy came close and fought the British. (The women who had stayed home when the British solders marched toward Lexington and Concord had been raped and watched their houses burn. The same thing happened in other areas. American militias had many women as a result.)

That was the first war when women in the United States fought. Before then, there was no United States.
