The attack on Fort Sumter by South Carolina militia and the cadets from the Citadel military school on April 12, 1861, was the actual beginning of the Civil War. John Brown's raid on Harper's Ferry a couple of years earlier is sometimes cited as the first salvo of the Civil War, but hostilities ended after Brown's followers were killed. Therefore the attack on Fort Sumter, which resulted in four years of non-stop warfare, is the best answer for the starting event of the Civil War.
