At the time they didn't particularly care and those who did just went ahead and settled where they wanted.
The proclamation of 1763 simply banned the colonists from settling west of the Allegheny Mountains. It was issued as an attempt to work out a peaceful agreement with the Native American tribes living in the west. The colonists, however, completely ignored the proclamation and continued attempting to colonize the west.
Nathaniel Bacon was a Virginia colonist most famous for instigating what is known as "Bacon's Rebellion" in 1676. He led other colonists in an armed rebellion against Governor William Berkeley for a variety of reasons, most immediately because the governor had prohibited the settlers from retaliating against the Native Americans for recent attacks on their settlements.

The original British colonies were east of the Appalachians. In fact, in the late 1750s, a law was passed that prohibited any settling beyond the Appalachians, even though the colonies extended beyond.
