In 1776...
The question is hard to answer because the US has been gaining land through conquest since the pilgrims landed. But if you mean when did it begin expand overseas, that would start with the annex of the Midway Islands in 1867, or with the purchase of Alaska and the Aleutian Islands in the same year.
The United States became a world economic power around the end of the 19th century, demonstrating its resulting military power through the Spanish-American War, among other adventures.
Around the time of World War 1, the United States became a world political power, exerting substantial influence on the treaties ending World War 1 and leading to the establishment of the League of Nations.
However, the United States did not fully accept its status as a world power until World War 2 and the outbreak of the Cold War.
1898, by the US capture of Spanish Empire possessions (the Philippines, Guam, Puerto Rico). At different times and using different methods, the US also acquired Alaska, the Hawaiian Islands, the Panama Canal Zone, US Virgin and Samoan Islands, and a stake in China, but by 1898 the US was definitely an empire.
The United States is not now, nor has it ever been, an "Empire". Such references are Propaganda spouted by those seeking to use the United States as a distraction and focus for their people away from the causes of their true suffering - their own governmental and religious repression.
Many examples of this exist in recent history: Hitler used the Jews, Saddam Hussein used the Kurds, in Africa other tribes were the scapegoats.
The U.S. in its history has always had territories - this is how the States originally were settled and came to be. Some territories have been given independence, other became States - the Phillippines and Hawaii are two examples. Puerto Rico, Guam and the U.S. Virgin Islands are example of territories that belong to the U.S., but where Statehood has not been accepted enough by the population. Each territory has its own government, with representation in the U.S.
Empires rule from a central government, and care little for the people in their control. The biggest example of why the U.S. is not an "Empire" is the amount of illegal Immigration the country deals with. The United States is a nation built by those fleeing repressive governments and looking for freedom. No one has ever tried to escape the United States to look for a better life elsewhere.
Since the US does not have colonies and it is not engaged in a program of global conquest, it is not really an imperialist country. However, the US does exert a lot of influence, and one might even argue, the US meddles in the affairs of many other countries, which is at least a somewhat imperialist kind of activity. All countries try to protect their own interests, and the more powerful a country is, the more effectively it can do so. Since the US is a very powerful country, so powerful that it is often called the world's only remaining superpower, following the collapse of the USSR, the US is in a position to throw its weight around.
The result of the Spanish/American War helped the United States become a world power by letting other countries know that the United States was not going to back down. Other countries did not want to start a conflict with the United States after this war.
