Prior to its work on the Declaration of Independence, the Second Continental Congress included Washington as a congressman from Virginia. But he resigned his position as a delegate when Congress formed the Continental Army and appointed him commanding general on June 14, 1775. So he was unavailable to participate, or to sign the Declaration.

Thomas Jefferson did not sign the Constitution, nor was he present at the Constitutional Convention of 1787 or when the First Amendment and religious freedoms were debated in the first session of Congress in 1789. During that time he was serving as US Minister to France (equivalent to Ambassador to France).

John Langdon did not sign the Declaration of Independence because he was not a delegate to the Second Continental Congress that debated and approved the document. He did serve as a delegate from New Hampshire to the Constitutional convention and signed the Constitution.

James Madison didn't sign the Declaration of Independence but he did sign the Constitution. He didn't sign the Declaration of Independence because it lacked a legitimate Bill of Rights.
